低温油脂降解菌的分离和菌剂构建及中试验证

Isolation of Low-temperature Oil-degrading Bacteria and Construction of Microbial Agent and its Pilot-scale Degradation Test

摘要: 通过从制革废水处理厂浓缩污泥池中分离筛选耐低温的土著油脂降解菌,采用部分因子设计法对所得菌株进行优化组合,制备了由4株高效油脂降解菌组成的固定化复合菌剂,并考察了中试条件下复合菌剂对制革含油废水的降解效果。结果表明:筛选获得了10株可在4 ℃下生长的油脂降解菌,其单菌种的混合油脂降解率最高可达75.36 %;对所得菌株Y3、Y7、Y9、Y10进行16S rDNA测序鉴定发现分别与苍白杆菌属(Ochrobactrum sp.)、芽孢杆菌属(Bacillus sp.)、伯克霍尔德菌属(Burkholderia sp.)有99%以上的一致性和高度进化同源性;复配混合菌制备为固定化菌剂,活化后投加至气浮生物反应器中,在接种量为2.5%(体积比),环境温度为13~15 ℃,处理时间为48 h的条件下,对体积分数为2.6%的含油制革废水油脂去除率可达86.1%。

Abstract: In this study, indigenous oil-degrading psychrotrophs were isolated from the concentrated sludge tank of tannery, and optimization and combination of these strains were performed by using the partial factor design method. The immobilized inoculants composed of 4 isolated strains was prepared and tested for oil degradability under low temperature on pilot-scale. The results showed that 10 strains of oil-degrading bacteria which can grow at 4℃were screened, and the maximum oil-degrading rate of single strain was up to 75.36%. 16S rDNA sequencing of the isolated strains Y3, Y7, Y9 and Y10 was conducted, and the results suggestedthat they were members of Ochrobactrum sp., Bacillus sp. and Burkholderia sp., respectively. The immobilized bacterial agents were prepared by the mixed bacteria, and the bacteria were added to the airlift bioreactor. Under the conditions of the inoculating amount of 2.5% (volume ratio), at ambient temperature of 13-15 ℃ for 48 h, the oil removal rate of the leather wastewater containing 2.6% (V/V) oil could reach 86.1%.
