典型牛皮中氨基多糖含量与分布及其在制革过程中的变化

Content and Distribution of Glycosaminoglycan in Typical Bovine Hides and its Variations with Leather Making Processes

摘要: 研究了典型牛皮中氨基多糖(GAG)含量及其在制革准备工段中随处理工序的变化。结果表明,黄牛皮和奶牛皮表皮层和粒面层中GAG含量高于网状层和肉面层,奶牛皮中的糖含量高于黄牛皮;在纵切面上,多糖含量沿表皮至肉面逐渐递减,在距离粒面约0.4 mm处GAG含量最高;PAS-AB特异性染色分析发现,在牛皮的表皮层和粒面层的基底膜、毛囊和腺体中含有较多的GAG,胶原纤维表面也观察到氨基多糖的分布。在制革准备工段中,皮内氨基多糖含量随处理工序的进行呈递减趋势,GAG的主要去除工序是浸灰和酶软化,常规制革准备工段中其去除率约为72%。皮中的GAG的去除与脱毛和纤维分散的效果紧密相关,需要进一步研究基于糖苷酶作用的安全、高效的GAG去除方法

Abstract: The content and distribution of glycosaminoglycan (GAG) in typical bovine hides and its variations with the beamhouse processes in leather making were studied. The results showed that the contents of GAG in the epidermis and grain layer of cattle and cow hide were higher than that in the reticular and flesh layer, and the content of GAG in cowhide was higher than that in cattlehide. The contents of GAG were reduced gradually from the epidermis layer to flesh layer, and on the layer of 0.4 mm to the grain surface shows the highest content of GAG. PAS-AB staining results showed that GAG mainly existed in the epidermis layer and the basement membrane, hair follicles and glands of the grain layer. The surface of collagen fibers also contains a certain amount of GAG. Additionally, the contents of GAG in the hides were gradually decreased with the conduct of beamhouse processes, especially the liming and enzymatic bating operations. The removal of GAG in leather making processes was closely related to the effect of unhairing and fiber opening-up, however, GAG removal ratio was only about 72% in the conventional beamhouse processes. Therefore, new developments in safety and efficient GAG removal methods based on the hydrolysis of glycosidase must have to be taken into consideration.
