耐铬厌氧胶原蛋白水解菌的筛选及应用研究

Screening and Application Research of Chromium-Resistant Anaerobic Collagen-Hydrolyzing Bacteria

摘要: 从制革厂含铬污泥中分离、纯化、筛选出一种耐铬优势真菌FN,经过梯度实验确定优势菌种对含铬胶原蛋白的耐铬质量浓度为1000 mg/L,并成功进行了厌氧驯化。真菌FN最终鉴定为米曲霉。将其用于含铬胶原蛋白的厌氧消解,考察了厌氧消解效果与产气性能。实验结果表明:耐铬菌接种量为25%、初始pH约为7,温度35 ℃,震荡110 r/min条件下,碳氮比为25时厌氧消化体系产气性能最佳,第二天达到最高日产气量295 mL,30 d累计产气1292.5 mL,甲烷含量52.57%,铬去除率为31.02%,蛋白质水解率为65.2%。碳氮比为35、初始pH约为7,温度35 ℃,震荡110 r/min条件下,耐铬菌投加量为50%时厌氧消化体系产气性能最佳,第二天达到最高日产气量414 mL,30 d累计产气1648 mL,甲烷含量45.18%,铬去除率为41.96%,蛋白质水解率86.49%。

Abstract: A chromium-resistant dominant fungus FN was separated, purified, and screened from the chromium-containing sludge in a tannery. Through the gradient experiments, it was determined that the dominant bacteria had a chromium-resistant concentration of 1000 mg/L for chromium-containing collagen, and the anaerobic acclimation of the strain was successfully carried out. The fungus FN was finally identified as Aspergillus oryzae. FN was then used in the anaerobic digestion of chromium-containing collagen, and the anaerobic digestion and gas production properties were investigated. The experimental results show that the inoculation amount of chromium-resistant bacteria is 25%; the initial pH is about 7; the temperature is 35 ℃, and the vibrating speed is 110 r/min. When the carbon/nitrogen (C/N) ratio is 25, the gas production performance of the anaerobic digestion system is the best. Next day, the highest daily gas production reached 295 mL. The cumulative gas production in 30 days was 1292.5 mL; the methane content was 52.57%; the chromium removal rate was 31.02%, and the protein hydrolysis rate was 65.2%. The C/N ratio is 35; the initial pH is about 7; the temperature is 35 ℃, and the chromium-resistant bacteria dosage is 50% under the vibrating conditions of 110 r/min. The gas production performance of the anaerobic digestion system is the best, and the highest daily output is reached the next day. The gas volume is 414 mL; the cumulative gas production in 30 days is 1648 mL; the methane content is 45.18%; the chromium removal rate is 41.96%, and the protein hydrolysis rate is 86.49%.
