胶原蛋白热变性检测方法的比较研究

Comparative Study on Detection Methods of Thermal Denaturation of Collagen

摘要: 采用圆二色光谱法、旋光度法、傅立叶变换红外光谱法、超灵敏差示扫描量热法以及X-射线衍射法对Ⅰ型胶原蛋白的热变性进行了检测,以探究胶原蛋白热变性不同检测方法的差异。结果表明,上述不同方法都是研究胶原蛋白热变性的有力工具,圆二色光谱法、旋光度法和超灵敏差示扫描量热法可以监测溶液中胶原蛋白的热变性过程,傅立叶变换红外光谱法和X-射线衍射法可以定性检测固体状态胶原蛋白的热变性。其中,超灵敏差示扫描量热法和圆二色光谱法与其他方法相比灵敏度更高。本论文结果将为胶原蛋白的相关研究提供参考。

Abstract: The thermal denaturation of type I collagen was studied by circular dichroism (CD), polarimetry, Fourier transform infrared spectroscopy (FTIR), ultrasensitive microcalorimetry (US-DSC) and X-ray diffraction (XRD). The results indicated that the above methods were the powerful tools to study the thermal denaturation of collagen. The process of thermal denaturation of collagen in solution could be monitored by CD, polarimetry and US-DSC. And the thermal denaturation of solid state collagen could be detected qualitatively by FTIR and XRD. Among them, US-DSC and CD were more sensitive than other methods. The results will provide a theoretical basis for the related research of collagen.
