Effect of Surfactant on Basic Properties and Leather Bating Performance of Trypsin

摘要: 文章考察了6种表面活性剂对胰蛋白酶基本性质的影响,结果表明阴离子型表面活性剂十二烷基硫酸钠会明显抑制胰蛋白酶活力,顺丁烯二酸二仲辛酯磺酸钠会显著增加胰蛋白酶溶液的粒径,均不适用于制革软化工序;十二烷基苯磺酸钠(SDBS)、脂肪醇聚氧乙烯醚硫酸钠(AES)、烷基酚聚氧乙烯醚(JFC-1)和脂肪醇聚氧乙烯醚(AEO-9)对酶活力影响小,且能大幅降低酶溶液的表面张力,但会略微增加酶溶液的粒径,其中阴离子型的SDBS和AES会增加酶分子的负电荷量。采用荧光示踪技术研究了SDBS、AES、JFC-1和AEO-9对软化过程胰蛋白酶在皮内传质行为的影响,结果表明这4种表面活性剂均有效提高了酶分子在裸皮中的传质速率,且表面活性剂对酶分子的粒径影响越小,强化传质的效果越好。使用表面活性剂有利于缩短软化时间,甚至减少酶用量。

Abstract: The effects of six surfactants on the basic properties of trypsin were investigated. The results showed that the anionic surfactant sodium dodecyl sulfate greatly inhibited the activity of trypsin, and di-sec-octyl maleate sodium sulfonate dramatically increased the particle size of trypsin solution. Therefore, the two surfactants are not suitable for the leather bating process. Sodium dodecyl benzene sulfonate (SDBS), sodium fatty alcohol polyoxyethylene ether sulfate (AES), alkylphenol polyoxyethylene ether (JFC-1) and fatty alcohol polyoxyethylene ether (AEO-9) had little effect on the activity of trypsin and drastically reduced the surface tension of trypsin solution, but they slightly increased the particle size of trypsin solution. Among the four surfactants, the anionic surfactants SDBS and AES could increase the negative charge of trypsin molecules. Furthermore, the effects of SDBS, AES, JFC-1 and AEO-9 on the mass transfer of trypsin in hide during bating process were studied by fluorescent tracing method. It was found that the four surfactants all obviously improved the transfer rate of trypsin in hide, and the surfactant that had less influence on the particle size of enzyme molecule enhanced the mass transfer more effectively. So, the use of surfactant in bating process is beneficial to shorten the bating time and even reduce the amount of trypsin.
