漆酶催化没食子酸改性胶原蛋白膜的研究

Modification of Collagen Membrane by Laccase-catalyzed Gallic Acid

摘要: 多酚与胶原主要以氢键发生相互作用。漆酶能将多酚氧化为醌,醌与胶原发生共价反应,交联效果更好。本研究利用漆酶催化没食子酸,使多酚和胶原发生共价交联以提升胶原膜的理化性能。通过单因素试验确定漆酶催化氧化没食子酸生成醌的最佳反应条件为:漆酶用量60 U、温度45 ℃、时间6 h、pH 5.5,反应液用于改性胶原膜。红外光谱证明漆酶催化没食子酸生成了醌式结构,且改性后胶原膜的三股螺旋结构保持完整。进一步实验表明,在漆酶的存在下,改性膜的力学性能、热稳定性和抗氧化性均得到了提升,而光透射率、溶胀率和水蒸气透过率均下降。因此,改性后的胶原膜可应用于食品包装行业,特别是肉类等易氧化变质的食物。

Abstract: Polyphenols interact with collagen mainly by hydrogen bonds. Laccase can oxidize polyphenols to quinones which can react covalently with collagen, leading to the better cross-linking effect. In this study, gallic acid catalyzed by laccase was used for covalent cross-linking with collagen to improve the physical and chemical properties of collagen membranes. The results of single factor test showed that the optimal reaction condition of laccase-catalyzed gallic acid could be obtained by using 60 U laccase at 45 ℃ and pH 5.5 for 6 h. The measurement of infrared spectrum indicated that laccase catalyzed gallic acid to quinone, and the triple helix structure of the modified membrane remained intact. Further experiments showed that in the presence of laccase, the mechanical property, thermal stability and antioxidant activity of the modified membrane were improved, while the light transmittance, swelling rate and water vapor transmittance were decreased. Therefore, the modified collagen membrane can be applied to the packaging of meat and other foods that are easy to be oxidized and spoiled.
