Effects of Water-based Solvents used for Collagen Electrospinning on the Structure of Fish Collagen

摘要: 研究了两类胶原电纺用水性溶剂体系(醋酸溶液和PBS/乙醇混合溶剂)对黑鱼皮胶原结构的影响。首先分析了文献报道水性溶剂体系的稳定性与表面张力,在此基础上分别用3个浓度的醋酸溶液(0.5 mol/L、1 mol/L和2 mol/L)及3个配比的PBS/乙醇混合溶剂(V(PBS)∶V(乙醇) 为3∶1、3∶0.5和3∶0.25)溶解鱼皮胶原制备胶原溶液,综合运用电泳、红外和紫外光谱以及超灵敏差示扫描量热等技术手段考察了不同鱼皮胶原溶液的结构性质。结果表明,两类水性溶剂体系随着醋酸或乙醇浓度的增加,都对鱼胶原的结构稳定性产生破坏,而且乙醇对鱼胶原的变性作用更强;醋酸溶液可能比PBS/乙醇混合溶剂更适合用于鱼胶原的连续稳定电纺,但有必要采取策略同步增强胶原的结构稳定性与静电可纺性。

Abstract: The effects of two kinds of water-based solvents used for collagen electrospinning (acetic acid solution and PBS/ethanol mixed solvent) on the structure of snakehead skin collagen were studied. Firstly, the stability and surface tension of the water-based solvents reported in literatures were analyzed. On this basis, three concentrations (0.5 mol/L, 1 mol/L, and 2 mol/L) of acetic acid solution and three types of PBS/ethanol mixed solvent (V(PBS)∶V(ethanol) equals to 3∶1, 3∶0.5, and 3∶0.25) were used to prepare fish collagen solutions. The techniques including electrophoresis, infrared and ultraviolet spectroscopy and ultra-sensitive differential scanning calorimetry were used to study the structural properties of different fish skin collagen solutions. Results showed that the two kinds of water-based solvents destroyed the structural stability of fish collagen with the increase of acetic acid or ethanol concentration, and the denaturation effect of ethanol on fish collagen was stronger. Acetic acid solution maybe more suitable for continuous stable electrospinning of fish collagen than PBS/ethanol mixed solvent, but it is necessary to take a strategy to simultaneously enhance the structural stability and electrostatic spinnability of collagen.
