Rheological, Thermal Properties and Chemical Crosslinking of Snakehead Collagen Solution

摘要: 研究了5 mg/mL黑鱼鱼皮胶原溶液的流变和热稳定性,并与相同浓度牛皮胶原溶液作比较;考察了温度对黑鱼胶原的动态粘弹性与成纤维过程的影响;对比分析了戊二醛和己二酸-N-羟基琥珀酰亚胺(NHS)酯用于交联改性黑鱼胶原溶液的效果。结果表明,相同浓度的黑鱼胶原与牛皮胶原溶液的流变学参数和热稳定性存在显著差别,黑鱼胶原的分子缠结程度较低、结构稳定性较差;黑鱼胶原的动态粘弹性与成纤维行为都具有明显的温度依赖性;己二酸NHS酯对黑鱼胶原溶液具有较好的交联改性效果,有望取代戊二醛用于提升鱼胶原基材料的结构性能。

Abstract: The rheological property and thermal stability of 5 mg/mL snakehead skin collagen (SSC) were studied and compared with bovine hide collagen (BHC) at the same concentration. The effects of temperature on the dynamic viscoelasticity and fibril formation of SSC were investigated. The effects of glutaraldehyde and adipic acid N-hydroxysuccinimide (NHS) ester on the crosslinking of SSC solution were studied. The results showed that there were significant differences in rheological parameters and thermal stability between SSC and BHC at the same concentration. The molecular entanglement degree was lower and the structural stability was poorer for SSC. The dynamic viscoelasticity and fibril forming behavior of SSC were obviously temperature dependent. The NHS ester had a good crosslinking effect on SSC solution, and was expected to replace glutaraldehyde to improve the structure and properties of fish collagen based materials.
