利用制革废弃牛毛制备角蛋白基水凝胶研究

Research on Keratin-based Hydrogel from Tanning Waste Bovine Hair

  • 摘要: 制革废弃牛毛是一种重要的角蛋白资源。采用甲基丙烯酸酐交联制革废弃牛毛还原角蛋白制备了甲基丙烯酰化角蛋白水凝胶。通过紫外分光光度计、FTIR、1H NMR及XPS等表征方法研究了甲基丙烯酸酐与还原角蛋白的化学交联机理。结果表明:还原角蛋白上的伯氨基可与甲基丙烯酸酐发生酰化反应,从而引入C=C双键,利用其与角蛋白分子自身结构中的巯基发生巯基-乙烯基反应,并且在中间酰化反应过程中产生的甲基丙烯酸副产物,也可与角蛋白中的巯基发生巯基-烯反应,从而得到三维网状结构,即可制备交联型甲基丙烯酰化角蛋白水凝胶。

     

    Abstract: Bovine hair waste in tannery is an important keratin resource. Methacrylated keratin hydrogels are prepared through crosslinking reaction between methacrylic anhydride and keratin extracted by reduction method from tannery bovine hair waste (K-SH). The reaction mechanism between methacrylic anhydride and K-SH has been studied by UV spectrophotometer, FTIR, 1H NMR and XPS. The results indicate that acylation reaction occur between the amino groups on the K-SH molecular chain and methacrylic anhydride to introduce vinyl groups, which can be reacted with thiol groups on the structure of K-SH via thiol-ene reaction. Meanwhile, methacrylic acid is formed during the acylation reaction, and it can also react with thiol groups on K-SH through thiol-ene reaction. Thereby, Methacrylated keratin hydrogels with three-dimensional network structure are obtained.
