胎牛皮源高层级胶原聚集体的制备与表征

The Controllable Preparation of Aggregated Collagen based on Traceable Fetal Bovine Skin

  • 摘要: 以实验室自制胎牛皮脱细胞真皮基质(FB-ADM)为原料,采用自主设计的可控降解技术制备了胶原(col)和超分子聚集态胶原结构体(高层级胶原聚集体,AG-col),流延成膜后,通过AFM、FT-IR、CD、DSC、TG、抗张强度检测、耐酶降解性能检测等手段对col和AG-col的性能结构进行表征,结果表明:二者红外特征吸收峰明显,三股螺旋结构完整,但AG-col的结构层次更高级,交联结构更加稳定,热稳定性(耐热破坏性和抗热分解能力)更好。此外,AG-col的强度、柔韧性和耐酶降解性能较col也更加优越。

     

    Abstract: In this study, by self-designed controllable-degradation method, collagen(col) and aggregated collagen(AG-col) were prepared from fetal bovine acellular dermal matrix (FB -ADM). Atomic force microscope (AFM), Fourier transform infrared spectroscopy(FTIR),circular dichroism (CD),differential scanning calorimetry(DSC), thermogravimetric analysis (TG), mechanical tests and collagenase degradation test were conducted to characterize col and AG -col. Results showed that both of them owned obvious characteristic peaks in FTIR spectra, indirectly demonstrating the exist of their characteristic triple helix structure. However, AG-col is more advanced than col, with better thermal stability. Furthermore, the mechanical property and resistance to enzymatic degradation of AG-col were also superior to col.
